Alcohol metabolism in the human body is regulated by two key enzymes: alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH). Genetic variations in these enzymes, particularly in the ADH1B and ALDH2 genes, significantly affect alcohol processing and health risks.

The ADH1B variant (rs1229984) increases enzyme activity, rapidly converting alcohol into acetaldehyde, a toxic intermediate. Meanwhile, the ALDH2 variant (rs671) reduces enzyme efficiency, slowing acetaldehyde breakdown. This leads to its accumulation, causing facial flushing, nausea, and a higher risk of esophageal cancer.
